706 Ceramide-1-phosphate accelerates premature skin aging in skin fibroblasts exposed to diesel particulate extract

چکیده
Extrinsic factors, such as air pollutants and ultraviolet irradiation (UVR), accelerate premature aging, a correlation between levels of skin aging symptoms has been reported. Diesel particulate matter, major source pollution, helps initiate some diseases, including diseases. We recently demonstrated that diesel extract (DPE) activates nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX), followed by activation sphingomyelinase (SMase), leading to ceramide (Cer) production in cultured normal keratinocytes (KC). characterized increases Cer-dependent lipid mediator induces apoptosis. Using human dermal fibroblasts (HDF), here we investigated whether/how DPE accelerates aging. Non-toxic concentrations increased cell senescence marker ß-galactosidase activity HDF. Following exposure, found mRNA/protein expression matrix metalloprotease (MMP)-1 MMP-3 activities, which are all associated with NOX was activated resulted Cer due SMase next asked whether or its metabolites (Cer-1-phosphate [C1P] sphingosine-1-phosphate [S1P]) promotes MMP-1/MMP-3 activation. inhibition kinase, but not sphingosine kinase suppressed activation, identified C1P via initiation an arachidonate pathway activating cPLA2, STAT1- STAT3-dependent transactivation. confirmed type 1 collagen degradation HDF DPE-mediated C1P-dependent pathway. is also only DPE, other external/internal stressors; i.e., UVR, cigarette smoke, inflammatory cytokines. Our evidence above points playing critical role response diverse stressors.

UVB-induced premature senescence of human diploid skin fibroblasts.
In this work, we show that repeated stresses with UVB (290-320 nm) induce stress-induced premature senescence (SIPS) of skin human diploid fibroblasts (HDFs). HDFs at early cumulative population doublings were exposed three or five times to increasing subcytotoxic doses of UVB with one stress per day. After 2 days of recovery, several biomarkers of replicative senescence were established. First...
متن کاملRole of Cultured Skin Fibroblasts in Aesthetic and Plastic Surgery
The skin is the largest tissue of man with several functions. Cosmetics/cosmeceuticals agents always are needed to be evaluated for their detrimental effects on the skin. Nowadays, the therapeutic potential of stem cells has and fibroblasts have increased the hope for a successful cell therapy in aesthetic medicine. Stem cells are unspecialized cells capable of renewing themselves through cell ...
متن کاملGadd45b deficiency promotes premature senescence and skin aging
The GADD45 family of proteins functions as stress sensors in response to various physiological and environmental stressors. Here we show that primary mouse embryo fibroblasts (MEFs) from Gadd45b null mice proliferate slowly, accumulate increased levels of DNA damage, and senesce prematurely. The impaired proliferation and increased senescence in Gadd45b null MEFs is partially reversed by cultur...
متن کاملEvaluating the Effect of First Skin to Skin Contact Father with Premature Infants Admitted to their Anxiety
Introduction: The birth of a premature baby shocks parents a lot. Although fathers also experience high levels of anxiety, they have received little attention and research. In order to focus on family care, this study was conducted to determine the effect of the first skin contact of fathers with premature infants hospitalized on their anxiety in the neonatal intensive care unit. Methods: This...
متن کاملA review on nutrition and skin aging
Background: Skin reflects the general health status and is not an exception in the process of aging. Intervention studies indicate that it is possible to delay skin aging and improve skin conditions through diet-based anti-aging strategies. The purpose of the current work was to review recent existing literature regarding the role of nutrition, for and against skin aging processes.Method: This ...
